Strictly, no, but they both serve to locate the disc, and to locate the wheel when refitting. Without, they're tricky to fit.

I don't have them and refitted the disc at 90 degrees to it's correct position. This gave me very spongy breaks.

Removed the disc and lined up with the correct holes, and this cured the spongy breaks.

Just a bit of a heads up for anyone I guess.

That... makes me wonder how, since everything should be flat.

However - if the stud has broken off close to the hub, as many have, with just a bit sticking out, then it is imperative that the disc be replaced with the holes over the stub. Otherwise, the pressure of the stub can deform the disc when the wheel is tightened over it. In my case it led to knocking brakes (and new discs to cure it) but I suspect that a smaller sticky-out bit might just increase the runout and push the brakes back into the callipers.

I think instead of the studs I have what I would describe as blanking nipples with a slight raise from the surface.

Then you need to make sure they align with the holes.

Aye, only realised this when I had my spongy brakes!

The one time I did it, when I took the disc off it had dimpled and cracked under the pressure.
